Onderzoekslijn Proliferative activity in hematologic disorders
Omschrijving Aims. Assessing proliferative behaviour by means of Ki-67 expression in tumour cells has high prognostic significance on survival and therapy response for many types of solid cancer. However, analysis of Ki-67 expression has never been explored in acute myeloid leukaemia (AML). This study aimed to investigate the fraction of Ki-67-positive cells within blood samples from AML patients to gain more insight into proliferative patterns found in this malignancy. Special emphasis was placed on Ki-67 expression in leukemic stem cells (LSCs),
which underlie the pathology of AML.
Methods. Multiparameter flow cytometry was used to identify hematopoietic stem cells (HSCs), leukemic stem cells (LSCs), multipotent progenitor cells (MPPs) and progenitor cells within bone marrow (BM) or peripheral blood (PB) of 28 AML patients. Within each population, Ki-67 expression was measured. Additionally, data were obtained from healthy femur head aspirates (n=25) to serve as a healthy control.
